China Develops Wheat Line with Stable Resistance to Fusarium Head Blight
A team of scientists in China has developed a novel wheat line called R111 that confers stable resistance to Fusarium head blight, a devastating fungal disease that can wipe out wheat harvests and poison grain. The researchers identified a quantitative trait locus (QTL) on chromosome 6A, which they named TaFhb-6A, as the source of this resistance.
The QTL was discovered using an integrated arsenal of genomic and transcriptomic tools, including bulked segregant exome sequencing (BSE-seq), non-denaturing fluorescence in situ hybridization (ND-FISH), and weighted gene co-expression network analysis (WGCNA). The team validated the locus by crossing R111 with susceptible wheat lines and genotyping the progeny using Kompetitive Allele-Specific PCR (KASP) markers.
The results showed that plants carrying the R111 allele at TaFhb-6A exhibited significantly enhanced FHB resistance, confirming that the locus is a genuine major contributor to the resistance phenotype. The researchers also identified two promising candidate genes within the QTL interval: TraesCS6A03G0005200 and TraesCS6A03G0009000.
Both genes are protein kinases involved in cellular signaling, with multiple missense mutations in R111 relative to susceptible parent MY11. The team found that both genes showed higher expression in resistant R111 than in susceptible MY11 after fungal inoculation, consistent with a faster or stronger immune signaling response in the resistant line.
